Interactions of AD risk alleles

   Proposal Description:
   Many different AD risk alleles have been identified but there is still very incomplete understanding of how they influence AD risk in combination with each other.  Some prior studies have looked at interactions and epistasis in AD risk alleles but more work is required in this area.  We intend to study interactions between the top several dozen (highest OR, lowest p-value will be the criteria for indicating the ?top? SNPs) risk alleles in terms of how they interact to confer altered risk for dementia according to neuropathologic criteria for disease operationalization.  We will obtain clinical and neuropathologic data from NACC and then query ADGC for the SNP data on the same cases.
